Abstract: Understanding cellular resource use is required for the predictive engineering of heterologous expression. In this study, the ribosome dynamics of E.coli were characterized under non-constant conditions and with/without heterologous GFP expression. It was observed that ribosome abundance and growth rate showed a cyclical relationship and that GFP expression did not depress ribosome abundance. A coarse-grain kinetic model was then built to recapitulate and explain the results.
