Title: | CELLULAR RESOURCE MODELS FOR GENETIC CIRCUIT DESIGN | Authors: | RUSSELL NGO JIE KAI | Keywords: | systems biology, synthetic biology, cellular resources, ribosomes, coarse-grained model, modeling | Issue Date: | 1-Jun-2022 | Citation: | RUSSELL NGO JIE KAI (2022-06-01). CELLULAR RESOURCE MODELS FOR GENETIC CIRCUIT DESIGN. ScholarBank@NUS Repository. | Abstract: | Understanding cellular resource use is required for the predictive engineering of heterologous expression. In this study, the ribosome dynamics of E.coli were characterized under non-constant conditions and with/without heterologous GFP expression. It was observed that ribosome abundance and growth rate showed a cyclical relationship and that GFP expression did not depress ribosome abundance. A coarse-grain kinetic model was then built to recapitulate and explain the results. | URI: | https://scholarbank.nus.edu.sg/handle/10635/230384 |
