dc.description.abstractThe present invention provides methods of immobilizing proteins onto a support, using a cellular expression system or a cell-free expression system, to attach a ligand to the C-terminus of the protein, by an intein-mediated or a puromycin-mediated approach. A method for improving the efficiency of intein-mediated ligand attachment is also provided. The methods of the present invention are useful in the preparation of protein microarrays.
