Abstract: Chondroitin sulfate C (CS-C) is widely used as a treatment option for osteoarthritis along with glucosamine but it has shown therapeutic potential in recovery and healing applications as well as having a dual role in the development, progression and metastasis of cancer. However, pure CS-C disaccharides and oligosaccharides are not easily accessible due to the difficulties in purification from natural sources and therefore chemical synthesis of CS-C is essential. This thesis describes the development of 2 synthetic methods for CS disaccharides and oligosaccharides (Building-Block Synthesis and Protecting-Group-Free Synthesis) and its effects on wound healing (cell proliferation, adhesion and migration) using HEPM cells as well as on breast cancer (cell proliferation) using MDA-MB-231, MCF-7 and BT-549. By designing and synthesizing a multi-functional chemical reporter that can be attached to CS, the CS chemical reporters could potentially be used for the mechanism of action of CS oligosaccharides on the biological system.
