Abstract: Resveratrol (RSV), a natural phytoalexin found in red wine has been shown to extend mean and maximum life span in organisms. Our study in F2 four-way cross hybrid mice evaluated the effects of ageing and long-term RSV treatment in drinking water for 6 or 12 months on biomarkers of oxidative damage and immunological responses. In the majority of tissues, there was a significant age-dependent accumulation of oxidative damage to DNA, lipid, protein and a clear increase in urine 8-Iso-prostaglandin2alpha (8-Iso-PGF2alpha) levels. Chronic RSV treatment elevated total RSV plasma levels and reduced age-dependent accumulation of 8-hydroxy-2'-deoxyguanosine in liver and heart; 8-Iso-PGF2alpha in heart and urine; and protein carbonyl content in liver and kidney. RSV intake ameliorated the age-related decline in phagocytic capability of granulocytes and T lymphoproliferation activity. The old RSV treated mice maintained cytokine profiles similar to that of a younger cohort with some pro-inflammatory and anti-inflammatory cytokines being elevated.
