Abstract: Die fill is an important step of pharmaceutical tablet production cycle. This research study attempts to critically define and understand several aspects of powder flow and die fill, and how they can impact mini-tablet production. Relationship between process parameters and die fill performance was found to be highly dependent on feed material properties and orifice diameter. Particle size and size distribution had affected non-cohesive arching and die fill performance. Various powder flow characterisation parameters were classified according to common physical attributes of the powders. Relationship between granule flow properties and die fill variation during mini-tablet production was dependent on the die fill mechanisms. Feed wheel paddle design and rotational speed had significant impact on mini-tablet die fill variation and tensile strength. Findings from this study could aid formulators in identifying and selecting critical powder attributes and process parameters in addressing potential die fill problems during mini-tablet production.
