Abstract: Based on our findings, I will explain the proposed calcium activation model of CapG, possibly applicable to other gelsolin family members as well. These findings explore how calcium activates CapG. This Intrinsic tryptophan fluorescence results suggest that CapG shares a similar calcium activation mechanism with gelsolin. Gelsolin uses its C-tail latch to regulate the calcium activation while CapG does not contain such a C-tail. The Quin2 titration along with NMR titration provide the binding affinities and order of calcium.The efforts to conduct a better understanding of calcium and PIP2 binding to CapG will also help to understand other gelsolins' activation mechanism.
