Abstract: DIVERSITY-ORIENTED FLUORESCENT LIBRARIES (DOFL) APPROACH IS ONE OF THE MOST POWERFUL METHODS TO DISCOVER NOVEL FLUORESCENT SENSORS. HERE, SEVERAL DYE LIBRARIES WERE SYNTHESIZED AND SENSORS FOR BIOTHIOL, NEURODEGENERATIVE DISEASE AND ELASTIN FIBERS WERE FOUND AND INVESTIGATED IN VIVO. BY CONSTRUCTING DARK DONOR WITH TUNABLE HIGH QUANTUM YIELD ACCEPTORS, A FRET BASED LIBRARY (BNM) WAS SYNTHESIZED. BNM COMPOUNDS SHOW SOME UNIQUE PHOTOPROPERTIES. THE MECHANISM WAS DEMONSTRATED BY ULTRAFAST DYNAMIC STUDY AND BASED ON THE THEORICAL STUDY, BNM BASED BIOTHIOL SENSOR WAS DESIGNED AND SYNTHESIZED. CELL BASED SCREENING USING DOFLA HAS BEEN PERFORMED AND A? OLIGOMER SPECIFIC SENSOR WAS DISCOVERED. THE FLUORESCENCE OLIGOMER SPECIFIC SENSOR SHOWS GOOD SELECTIVITY IN VITRO AND CAN SHOW CLEAR A? OLIGOMER STAINING IN THE BRAIN TISSUE. A HIGHT PHOTOSTABILITY ZWITTERIONIC LIBRARY, CYZW, WAS ALSO DESIGNED AND SYNTHESIZED FOR BIOIMAGING IN VIVO. AN ELASTIN FIBERS PROBE CYZW-H5 WAS FOUND BY S
