dc.description.abstract | Fluorescence correlation spectroscopy (FCS) and its modality fluorescence cross-correlation spectroscopy (FCCS) are single molecule sensitive optical tools to study mobility, concentrations and interactions. The aim of this thesis is to study the interaction, diffusion and distribution of epidermal growth factor receptor (EGFR) within the plasma membrane through two variants of FCS, single-wavelength-FCCS (SW-FCCS) and total internal reflection FCS (TIR-FCS). EGFR is a tyrosine kinase that plays a fundamental role in a large variety of physiological and pathological processes in cells. The quantitative analysis of EGFR dynamics provides new information on its functioning process. Here I analyze the homodimerization of EGFR before activation, the binding between EGFR and a phosphotyrosine binding domain (PTB) after stimulation, the suppression of the binding by a EGFR-specific phosphorylation inhibitor, and the EGFR diffusion and distribution on the plasma membrane in dependence of cholesterol concentration, the cytoskeleton and the activation. | |
