Abstract: In this thesis, a series of p-conjugated materials featuring star-shaped donor-acceptor architectures, using [2,2]paracyclophane, triphenylamine or pyrene as the core was developed for theoretical study and for two-photon microscopy (TPM) bioimaging applications. Star-shaped structures with different donors and acceptors were successfully synthesized with large two-photon absorption (TPA) cross sections and tunable emission from blue to red in water. Moreover, molecular engineering strategies using sugar moieties were also developed for enhanced TPA cross sections and a targeting functionality of water soluble TPA materials. Three of the materials have been successfully demonstrated for two-photon fluorescence cellular imaging in a high contrast manner. In summary, the synthetic methodologies, the donor-acceptor systems, the molecular engineering strategies demonstrated as well as the underlying mechanisms unveiled in this PhD study provide useful guidelines in future advancement of TPA materials.
