dc.description.abstract | As a part of the effort to investigate liver and pancreas development in zebrafish, this thesis focused on the function of rbp4 (retinol binding protein 4) in early liver development and the cell lineage relationship in pancreas. rbp4 expression was restricted to the yolk syncytial layer before liver bud formation. Knockdown of rbp4 resulted in liver bud duplication presumably due to the impaired migration of liver precursors. To study pancreas cell lineage, genetic ablation by transgenic expression of diphtheria toxin gene A was employed. A stable transgenic line was created to target GFP expression in somatostatin cells. By combination of other pancreas-specific GFP transgenic lines, we ablated both exocrine and endocrine cells and found that endocrine and exocrine cells were developed independently. Meanwhile, ablation of some endocrine cells led to defects in other endocrine cells. Based on our observations, a tentative model for endocrine cell lineage is presented. | |
