Abstract: This thesis systematically explored the structural features (backbone bending, columnar packing and potential channel formation) of conformationally stabilized crescent, helical and circular aromatic oligoamides. Except for exhaustive structural studies, the application of aromatic oligoamides as G-quadruplexs ligands was also studied by traditionally methods and a novel efficient gold nanoparticle screening assay. Firstly, the structure of crescent folding backbones of oligoamides was explored and the location-dependent strength of multiply centered intramolecular H-bonding was assessed. Also, the structural and physical properties of helical conformation enforced by the continuous hydrogen-bonding in foldable aromatic oligoamides were investigated. Then, pentagon-shaped circular aromatic oligomers were synthesized and the binding affinities of aromatic oligoamides towards G-quadruplexs were studied. Finally, a novel and efficient aptamer-based gold nanoparticle (AuNPs) assay was designed for screening G-quadruplexs ligands based on the exhaustive studies of interaction between thrombin and its G-quadruplxes aptamer.
