dc.description.abstract | Vascular dementia (VaD), the second most common cause of dementia worldwide, is a progressive neurodegenerative disorder that leads to cognitive decline. The unknown pathophysiology of VaD underpins the difficulty of targeting the root cause of the problem. Given the lifestyle-associated risk factors for VaD such as diabetes and hypertension, the environment seems to play a key role in modulation of gene expression. The gene-environment interaction (i.e. epigenetics) is hence hypothesised to explain the underlying mechanistic changes observed in the pathophysiology of VaD. This research thus explores the role of DNA methylation (an epigenetic mechanism) in a mouse model of VaD. In addition, it also introduces intermittent fasting (IF), a form of time-restricted feeding, as a prophylactic intervention. The findings of this research is pivotal in demonstrating the promise it holds in possibly slowing down the disease progression of VaD and conferring potential beneficial effects in the context of DNA methylation. | |
