dc.description.abstract | In plants parental genomic imprinting relies primarily on DNA methylation by the METHYLTRANSFERASE MET1, which is maintained from the silenced allele and removed from the active allele in a parent-specific fashion. Here we show that the RNA directed DNA methylation (RdDM) pathway orchestrates parental genomic imprinting at two loci that produce small interfering RNAs (siRNAs). At these loci, RdDM silences expression of the imprinted genes in vegetative tissues and male gametes by de novo DNA methylation. The maternal allele is activated by DNA demethylation in female gametes, resulting from the transcriptional inactivation of the essential RdDM component NUCLEAR RNA POLYMERASE D2 (NRPD2) and the action of demethylase DEMETER1. The results suggest that a new class of RdDM-dependent imprinted genes remain to be explored in Arabidopsis and our findings explain how imprinted expression could evolve from insertion of transposons producing siRNAs. | |
