dc.description.abstract | This thesis presents the optimization of the line scan focal modulation microscopy(LSFMM) system in terms of theoretical studies, system optimization, and combination with other novel imaging techniques. Firstly, an in-depth point spread function (PSF) mathematical model was built up to evaluate the imaging performance of the LSFMM system by computational simulation. Aberration effect was discussed to further optimize the optical system design. Secondly, a miniatured LSFMM system was proposed to be more compact and flexible for imaging studies on cells and tissues. Lastly, we tapped into the potential of LSFMM by combining with other innovative techniques such as structured light illumination and pixel reassignment to further improve the optical sectioning capability and image resolution. | |
