dc.description.abstractTobacco mosaic virus (TMV) is the most studied model for host-virus interaction in plants. It is a positive, single-stranded RNA virus. It encodes two replicases (126kDa and 183kDa), a movement protein and a coat protein. These proteins interact with many host proteins for virus replication. The first aim was to dissect the proteome profile of TMV-infected Nicotiana tabacum plant by using iTRAQ technique. We identified important host proteins involved in photosystems and plant defence (thioredoxin H-type1). Further, we identified that TMV-43A (TMV mutant) could cross-protect N. benthamiana plants against TMV using the cross-protection technique. We found DXR, GGPS and thioredoxins were crucial for cross-protection. Moreover, the third aim was to identify interacting or non-interacting host proteins to TMV replicase by using the novel BioID technique. We identified NbSGT1 protein which suppresses TMV replication by interacting indirectly to TMV replicase. These obtained results offer an important foundation for future agricultural applications.
