Abstract: The patterning of zebrafish somite involves participations of multiple morphogens at district body axis and rapid cell migrations. However, how cell differentiations are coordinated with cell migrations remains poorly understood. In this study, we utilized whole-somite 4D live imaging and cell tracking in combination with multiple genetic perturbations to systemically reveal cell differentiation and migration with zebrafish somite. Our data demonstrated that somite patterning and myotome formation are underpinned by a series of highly organized and directional cell morphogenesis and cell migration events. FGF imposes tight temporal control on the Shh/BMP activation/repression of slow muscles through regulating the time at which fast-twitch progenitors displace slow-twitch progenitors from contacting the Shh-secreting notochord. Last, we explored interactions between different somitic populations: the myogenesis of fast muscle progenitors is required for the lateral migration of slow muscle; the migrating slow muscle fibres reciprocally regulate the myogenesis and morphogenesis of the fast muscle fibers.
