dc.description.abstractThe re-emergence of Enterovirus 71 (EV71) as a neuroinvasive virus in the Asia Pacific region has led to increasing reports of fatal neurological complications in infected young children. With no effective drugs or vaccine to date, elucidating the mechanisms involved in EV71 neuropathogenesis has become of outmost relevance and importance. To gain insights into the invasion process of EV71 into the central nervous system, we have established a novel in vitro infection model using the motor neuron cell line NSC-34. EV71 infection cycle in this cell line was characterized and led to unique observations related to entry, replication and egress mechanisms employed by the virus. Furthermore, a proteomic approach was undertaken to map the dynamic host protein regulation during EV71 infection in NSC-34 cells. We identified prohibitin (PHB) as the first host factor that is specifically involved in EV71 neuroinvasion and a potential drug target to limit neurological complications.
