Abstract: Cancer is among the leading causes of death in the world. Some cancer cells are known to have dysregulated expression of the osteoarthritis causing protease aggrecanase-2 or ADAMTS5. Previous work from our lab showed anti-cancer activity of this protease in mice when overexpressed in cancer cells. The anti-cancer activity of ADAMTS5 was mapped to the central TSR domain (TSR1). However, it is unclear if physiological ADAMTS5 can also suppress metastasis. In this work, I investigated the role of physiological ADAMTS5 in metastasis using Adamts5-/- mice and characterized the metastatic properties of ADAMTS5-overexpressing tumour cell in culture. Furthermore, I explored the potential of the recombinant TSR1 as a potential cancer drug. My results suggest that host physiological ADAMTS5 promotes lung metastasis of melanoma in mice and ADAMTS5 confer high metastatic potential on melanoma cells.
