dc.description.abstract | Umbilical cord blood (UCB) is an allogeneic source of hematopoietic stem and progenitor cells (HSPC) for performing hematopoietic stem cell transplantation (HSCT) to correct disorders of the bone marrow (BM). The use of UCB grafts in adult patients is limited by the low cell dosage of both total nucleated cells (TNC) and HSPC. Successful ex vivo expansion strategies overcome low cell content while maintaining the repopulation functionality of the manipulated graft. This thesis aimed to perform clinically relevant ex vivo expansion of UCB grafts without the need to perform prior stem cell enrichment, using nanomaterials like surface functionalized single walled carbon nanotubes (f-SWCNT) and novel azole-based small molecules. The effects of the expansion platforms were analyzed using molecular, phenotypic and in vivo functional assays. The findings reported hereby establish carboxylic acid functionalized SWCNT and a novel azole-based small molecule C7 as regulator of HSPC fate and expansion. | |
