dc.description.abstract | Tubercle bacilli are obligate aerobes but have the ability to adapt and survive hypoxia by shifting down to non-repilcative drug resistant dormant form. Hence, hypoxic dormant bacilli could contribute to the observed persistence of the pathogen during chemotherapy. Temporal proteomic analysis of the dormancy response showed a drastic up-regulation of the alpha-crystallin homolog, a putative response regulator Rv3133c and conserved hypothetical proteins Rv2623 and Rv2626c in dormant bacilli. Importantly, the induction of Rv3133c was specific to the dormancy response. Disruption of the gene encoding Rv3133c results in a drastic loss of viability and loss of induction of the other three dormancy induced proteins. This provides convincing evidence that Rv3133c is the key regulator of the mycobacterial dormancy response. As such we named the Rv3133c gene dosR for dormancy survival regulator. | |
