dc.description.abstractCurrent methods for obtaining fetal cells for prenatal diagnosis are invasive and carry a small risk of fetal loss. Recovery of first trimester fetal erythroblasts from maternal blood represents an attractive alternative for non-invasive prenatal diagnosis. However, these cells are rare and it is challenging in recovering them from maternal blood due to the lack of surface markers for isolating fetal erythroblasts from adult red blood cells (RBCs).This thesis investigated the membrane proteins of fetal erythroblasts and adult RBCs using proteomic approaches and immunocytochemical screening. Twenty-six membrane proteins were found to be differentially/uniquely expressed. Of these, CD147 was demonstrated to be very useful for separating fetal erythroblasts from adult RBCs in model mixture. I envisage that this antigen would be useful for enhancing current enrichment protocol of isolating fetal NRBCs from maternal blood in the first trimester of pregnancy for an early non-invasive prenatal diagnosis.
