Abstract: BONE MARROW REPRESENTS AN ALTERNATIVE BONE REPAIR THERAPY TO AUTOLOGOUS BONE GRAFTS (ABG), DUE TO ITS OSTEOGENIC CELLS AND OSTEOINDUCTIVE GROWTH FACTORS. HOWEVER, THE EFFECTIVE USAGE OF BONE MARROW ASPIRATE (BMA) IN ORTHOPAEDICS ARE INCONSISTENT. PREVIOUSLY, WE REPORTED THE DEVELOPMENT OF AN AFFINITY-SELECTED HS VARIANT THAT BINDS TO BMP-2 (HS3+VE) AND INCREASED ITS STABILITY AND BIOACTIVITY. HERE, WE HIGHLIGHTED THE INTER-INDIVIDUAL VARIABILITY OF BMA CONSTITUENTS, PARTICULARLY THAT OF BMP-2 LEVELS. FURTHERMORE, HS3+VE IS ABLE TO BIND TO ENDOGENOUS BMP-2 IN RABBIT BMA IN VITRO. WHEN USED IN CONJUNCTION WITH AUTOLOGOUS BMA IN RABBIT SEGMENTAL ULNA DEFECTS, HS3+VE ENHANCED BMA?S REPARATIVE EFFECTS AND RESULTED IN MORE SUPERIOR AND CONSISTENT THERAPEUTIC OUTCOMES WHEN COMPARED TO ABG. MOREOVER, HS3+VE BOOSTED THE IN VIVO PERFORMANCES OF BMA IN THE PRESENCE OF 2 POLYCAPROLACTONE-BASED BIOMATERIALS. ERGO, HS3+VE IS A POTENT NON-PROTEINACEOUS NATURAL ADDICTIVE THAT PRESERVES NATIVE REPARAT
