dc.description.abstractTHE MULTI-SUBUNIT ENZYME COMPLEX F1FO ATP SYNTHASE IS A KEY ENZYME IN CELLULAR ENERGY METABOLISM. IN MYCOBACTERIUM THIS ENZYME CONSISTS OF A MEMBRANE-EMBEDDED FO DOMAIN AND A CYTOSOLIC F1 PART. SUBUNIT ? IS PART OF THE F1 DOMAIN AND LINKS IT WITH THE FO DOMAIN. IT CONTAINS A UNIQUE AMINO ACID SEQUENCE INSERTION, ?166-179, WHICH IS NEITHER PRESENT IN EUKARYOTES NOR IN OTHER PROKARYOTES. THE FUNCTION OF THIS SHORT PEPTIDE INSERTION IS UNKNOWN. USING RECOMBINEERING TECHNIQUE, I CREATED A MUTATION IN THE M. SMEGMATIS MC2 155 GENE ATPG CODING FOR SUBUNIT ? OF F1FO ATP SYNTHASE LACKING A 42 BP DNA SEQUENCE CODING FOR THIS LOOP. COMPARATIVE GROWTH PHYSIOLOGY STUDIES ON A WHOLE CELL LEVEL UNDER AEROBIC AND ANAEROBIC CONDITIONS DID NOT REVEAL ANY DIFFERENCES BETWEEN M. SMEGMATIS WILD TYPE AND ?ATPG166-179 MUTANT.
