dc.description.abstract | Quantitative investigation is important during liver fibrosis study by measuring, quantifying and locating variations of collagen and cellular structure changes. Two-photon excited fluorescence (TPEF) and second harmonic generation (SHG) are established label-free techniques to obtain such information. However, conventional multiphoton microscopy is limited by frame rate. This can be improved by using multifocal multiphoton microscopy (MMM). In this thesis, the correlation of forward and backward SHG signals during fibrosis progression was investigated. A more accurate diagnosis was achieved by combined features. To realize fast speed imaging, an integrated imaging system composing of both MMM and SHG techniques was established to enhance imaging speed by 64 times. An image post processing method for emission photon reassignment was developed to improve image contrast. Coupled with an automated slicing module, large volume tissue samples can be imaged at a high speed without information loss for liver fibrosis study in 3D. | |
