dc.description.abstractAnterior cruciate ligament (ACL) injury is common in athletes. Tissue engineering method is used to regenerate the ACL, but it faces the problem of missing the bone-ligament interface, which is an important structure made up of bone, fibrocartilage and ligament. Fibrocartilage is further subdivided into calcified fibrocatilage and uncalcified fibrocatilage, contributing to the smooth transfer of mechanical force in ACL. In this thesis, the bone-ligament interface was engineered by making use of the multi-potency of bone marrow stem cell as well as the different bimolecular cues loaded composite silk fibroin scaffolds. In more details, the BMSC was pre-differentiated into tri-lineage(i.e, osteogenic, chondrogenic and fibroblastic) on individual scaffolds as well in individual induced medium for 1st three weeks, and then the tri-lineage pre-differentiated BMSC were integrated into one scaffold complex by suturing and co-cultured under the common medium for another three weeks, finally forming a bio-mimic transitional bone-ligament interface.
