Abstract: THIS STUDY EVALUATED THE ASSOCIATION BETWEEN Y BOX BINDING PROTEIN (YB-1) AND GASTRIC TUMORIGENESIS. IMMUNOSTAINING OF YB-1 IN GASTRIC CANCER TISSUE MICROARRAYS WAS FOUND TO BE SIGNIFICANTLY CORRELATED WITH LAUREN CLASSIFICATION AND LYMPH NODE STATUS IN ALL TYPES OF GASTRIC CANCER. IN VITRO STUDIES PERFORMED, REVEALED THAT KNOCKDOWN OF YB-1 IN NUGC3 DIFFUSE GASTRIC CANCER CELLS, SIGNIFICANTLY INHIBITED CELL PROLIFERATION AND MIGRATION PROCESSES BUT NOT INVASION, WHEREAS DOWN-REGULATION OF YB-1 IN MKN7 INTESTINAL GASTRIC CANCER CELLS, IMPAIRED CELL MIGRATION AND INVASION. UP-REGULATION EFRRI1 GENE, OF A NEGATIVE REGULATOR OF EGFR KNOWN TO AFFECT THE MAPK PATHWAY, WAS IDENTIFIED BY MICROARRAY ANALYSIS. THE GENE EXPRESSION OF MAPK PATHWAY MEMBERS, MAPK1, MEK5 AND RSK WERE FOUND SIGNIFICANTLY DECREASED IN YB-1 SILENCED NUGC3 CELLS WHEREAS OBVIOUS DECREASE OF PROTEIN EXPRESSIONS IN P-ERK, P-EGFR AND P-RSK WERE ALSO REVEALED BY WESTERN BLOT ANALYSIS.
