dc.description.abstract | ELKS WAS PREVIOUSLY IDENTIFIED AS AN ACTIVE ZONE PROTEIN WITH IMPORTANT ROLES IN NEUROTRANSMITTER RELEASE, PRESUMABLY BY COOPERATING WITH MULTIPLE SYNAPTIC PROTEINS TO MODULATE SYNAPTIC VESICLE EXOCYTOSIS AT PRESYNAPTIC SITES. ELKS WAS RECENTLY SUGGESTED TO BE INVOLVED IN REGULATING INSULIN SECRETION. HOWEVER, THE MOLECULAR MECHANISM AND FUNCTIONAL ROLE OF ELKS IS NOT CLEARLY UNDERSTOOD. WE GENERATED A PANCREAS-SPECIFIC ELKS1/ELKS2 KO MOUSE LINE AND FOUND THAT THESE MICE EXHIBITED SEVERE GLUCOSE INTOLERANCE, IMPAIRED INSULIN SENSITIVITY, MILD HYPERGLYCEMIA AND HIGHER CIRCULATING INSULIN LEVELS. INSULIN SECRETION IN THE KO ISLETS WAS SIGNIFICANTLY IMPAIRED IN BOTH 1ST AND 2ND PHASES, AS A RESULT OF REDUCED INSULIN GRANULE DOCKING AND PRIMING. RESCUE EXPERIMENTS REVEALED THAT INTERACTIONS AMONG ELKS, RAB GTPASE AND RIM ARE IMPORTANT IN INSULIN GRANULE DOCKING AND PRIMING, RESPECTIVELY. MOREOVER, ELKS KO ?-CELLS DISPLAYED LOWER CA2+ CHANNEL ACTIVITY IN GLUCOSE-STIMULATED CONDITIONS. WE D | |
