Abstract: MULTIPLE C2 DOMAIN AND TRANSMEMBRANE REGION PROTEIN 1 (MCTP1) CONTAINS TWO TRANSMEMBRANE REGIONS AND THREE C2 DOMAINS OF HIGH BINDING AFFINITY FOR CA2+. ALTHOUGH A SINGLE-NUCLEOTIDE POLYMORPHISM (SNP) OF HUMAN MCTP1 GENE IS REPORTEDLY ASSOCIATED WITH BIPOLAR DISORDER, EXPRESSION AND FUNCTION OF MCTP1 IN THE CENTRAL NERVOUS SYSTEM IS STILL LARGELY UNKNOWN. WE CLONED MCTP1 ISOFORMS FROM THE RAT BRAIN. IN SITU HYBRIDIZATION HISTOCHEMISTRY (ISH), IMMUNOHISTOCHEMISTRY (IHC) AND IMMUNOELECTRON MICROSCOPY (IEM) REVEALED MCTP1 MAINLY IN NEURONS OF THE LIMBIC SYSTEM, PARTICULARLY IN SYNAPTIC VESICLES OF EXCITATORY SYNAPSES IN THE RAT BRAIN. IN CULTURED DEVELOPING NEURONS AND PC12 CELLS, MCTP1 IS LOCALIZED IN ENDOCYTIC RECYCLING PATHWAY. TF488 UPTAKE ASSAY REVEALED THAT OVEREXPRESSION OF MCTP1S IN PC12 CELLS SIGNIFICANTLY INHIBITED THE ENDOCYTOSIS OF RECYCLABLE CARGOES FROM PLASMA MEMBRANE. OVEREXPRESSION OF MCTP1S ALSO RETARDED THE RETRIEVAL OF SECRETORY VESICLES, SLOWED CELL MIGRATION, AND PR
