Abstract: ECM samples were modified by embedding streptavidin coated super-paramagnetic beads that form strong covalent bonds with the ECM fibers. The micro and macro scale stiffness of the modified ECM were tested and verified. Observations on cellular resposnse to the modified ECM were also carried out. The work reported in this thesis provides a set of fundamentally significant results that can serve as a point of departure for further study in active manipulation of ECM stiffness. By controlling the stiffness of the ECM, it may be possible to influence many biological processes such as cell migration, angiogenesis, stem cell differentiation, lymphangiogenesis and metastasis.
