Abstract: This thesis described the design and synthesis of macrocyclic pentamers and helical oligomers and their applications in stabilizing telomeric DNA and RNA G-quadruplex in both Na+ and K+ solutions. The G-quadruplex structure presents a novel structural feature where a planar G-tetrad comprises four in-plane guanine residues with an electron-rich p-surface. And the stabilization of telomeric DNA and RNA has a critical effect on the telomere related project such as anticancer drug discovery. Compounds with interior tunable functional groups or exterior amine terminated linkers were synthesized. In this thesis, circular dichroism (CD) was used to examine the Tm value of different sequences in the absence and presence of ligands. Some of these compounds presented very good selectively stability of human telomeric RNA G-quadruplex (UUAGGG)n rather than human telomeric DNA G-quadruplex (TTAGGG)n both in Na+ and K+. All these compounds have no binding with DNA duplex.
