Abstract: A recent advancement of the fluidized bed processor was the introduction of a FlexStreamTM processor module with swirling airflow in place of the typical axial airflow module of the conventional fluidized bed processor. This new processor is the main focus of this study. Design of Experiment (DOE) approach was employed to understand and optimised the granulation process. Important parameters were examined in the study and correlated to the qualities of the granules produced. FlexStreamTM granules produced were compared to top-spray granules using different spray rates. FlexStreamTM granules had better qualities at a high spray rate. In order to use FlexStreamTM processor for small beads drug layering, coating formulations with 2 grades of hydroxypropylmethyl cellulose (HPMC), HPMC E3 and HPMC VLV respectively, were compared. HPMC VLV was a better choice and coupled with optimal spray rate; uninterrupted drug layering of small beads to one to one weight gain could be achieved.
