Abstract: This thesis deals with the identification of specific allergens from major allergy-causing mites and the IgE binding patterns in several atopic populations. The study is compartmentalized into two main sections, the first describing the underlying IgE binding patterns of nine different species using sera from different populations; and the second focusing on the isolation of these allergens for subsequent analyses.For this purpose, an Expressed Sequence Tag (EST) collection of the storage mite Glycyphagus domesticus was produced, and its constituent allergens isolated. Comparative analysis was performed at the molecular level with corresponding homologues from major mite species such as Blomia tropicalis and Dermatophagoides spp. The results reported here would contribute significantly towards the prioritizing of candidate allergens for diagnostic regimes, as well as the identification of specific determinants for subsequent development of immunotherapeutic vaccines.
