Abstract: A series of novel self-assembled core-shell nanostructures were developed based on star polyrotaxanes, consisting of star PEG, alpha-CD, end blocking group with biodegradable linkage and cationic polymers. In the field of drug delivery, in order to meet the conflicting requirements of an ideal drug delivery system, disulfide-linkage was introduced. The nano-particles exhibited sustained and reductive in vitro drug release, which accomplished much faster internalization, rapid intracellular release of DOX and higher anticancer efficacy. Besides drug delivery, reduction-sensitive polyrotaxanes was developed as gene carriers. The systems could achieve rapid intracellular release, effective unpacking ability, low cytotoxicity, high transfection efficiency and high stability in biological fluids. It could also be very promising in delivering anticancer drug and gene simultaneously for improved cancer therapy. In sum, this research is helpful in creating a new approach to the design of reduction-sensitive delivery system which could be applied in drug and gene delivery.
