ScholarBank@NUS Repository. | Abstract: | Lymphangiogenesis is the formation of new lymphatic vessels from pre-existing vasculature, where lymphatic vessels are important in the maintenance of tissue fluid homeostasis and immune surveillance. Recently, we found that immunization with a strong adjuvant induces lymph node swelling and lymphangiogenesis in these inflamed lymph nodes. This process requires the recruitment of B cells in the lymph nodes. However, the signaling pathway underlying lymphangiogenesis in this inflammatory model has not been identified. We hypothesize that lymphotoxin B receptor signaling mediated by B cells may drive this inflammation-induced lymphangiogenesis. Our findings reveal that indeed, inhibition of lymphotoxin B receptor signaling blocked lymph node swelling and lymphatic vessel network expansion at the early stage of the process and involves the expression of matrix metalloproteinase-13. The proteolytic activity of matrix metalloproteinase-13 is important for the degradation of the extracellular matrix and basement membrane to enable the sprouting of new lymphatic vessels in vitro. In all, we showed for the first time the regulation of matrix metalloproteinase by lymphotoxin B receptor signaling in mediating lymphangiogenesis. | URI: | http://scholarbank.nus.edu.sg/handle/10635/38816 |
