dc.description.abstract | Regulation of cell cycle is crucial for cell survival and function. Dysregulated cell division can cause many diseases, including cancer. How cellular lipid changes affect the cell cycle and vice versa have become an increasingly important question. Here, lipid composition of cells in different cell cycle stages was studied using mass spectrometry-based lipidomics approaches. Two complementing synchronization methods (aphidicolin and nocodazole) were applied on two cell lines (HeLa and MCF7). In terms of overall cellular lipid composition, all lipid classes, except cholesterol esters, did not show significant changes in the cell cycle. Levels of cholesterol esters measured were generally higher in G2/M cells. Prolonged G2/M phase in ACAT1 knocked-down cells which had lower amount of cholesterol esters supported the observation. Cholesterol esters levels were also found to be higher in breast tumour tissues compared to control. Overall, our findings indicate that cholesterol esters are crucial in G2/M progression and possibly, tumorigenesis. | |
