Abstract: Gibberellins are the key phytohormones promoting germination, via RGL2, the key DELLA protein regulating this response. Functional and stable RGL2 is sufficient to inhibit germination completely by unknown molecular mechanisms. We investigated events directly downstream of RGL2, analysing the seed transcriptome differentially regulated by RGL2 and identifying over-represented cis-regulatory motifs present in promoters of genes affected. Our results indicate that RGL2 causes seeds to enter a state of dormancy. Several promoter motifs are enriched in promoters of differentially regulated genes. Of these, we confirmed the involvement of Dof-bound motifs in RGL2-mediated transcriptional regulation in an inducible reporter gene assay in protoplasts. By Chromatin Immunoprecipitation (ChIP) we confirmed selected genes to be directly targeted by RGL2, and characterised germination defects in mutants of selected target genes. Lastly, the MADS-box transcription factor AGL67 was analysed in more detail. Its expression is regulated by RGL2, and seeds of loss-of-function mutants are less dormant.
