Abstract: Breast cancer is the second leading lethal cancer type among females. Its highly invasive and metastatic features are among the major contributors to the associated high mortality rate. Therefore, compounds with anti-invasive and anti-metastatic activities might be effective therapeutic agents for treatment of this cancer type. Chalcones are a class of compounds bearing a 1,3-diphenyl-2-propen-1-one structural backbone and have been reported to possess anti-inflammatory, anti-cancer, anti-oxidant and anti-infectious effects. A number of naturally-occurring chalcones have been reported to possess anti-invasive effects against several tumor types. However, the underlying mechanisms remain fully elucidated. In this present study, a series of chalcone analogs containing hydroxyl, fluoro and methoxy substitutions were evaluated for potential anti-invasive effects against breast carcinoma cell lines of invasive phenotype. Analogs with potent anti-invasive effects were identified and the underlying mechanisms were further investigated.
