Abstract: Recent findings suggest that there exists an alternative PI3K-Akt independent survival pathway in LNCaP. Increasingly, there are evidence suggesting that oxidative stress is correlated to cancer aggressiveness and increased chemoresistance. In this project, the parameters influencing LNCaP survival was defined, and used to investigate the mechanism of cell survival induced by serum and superoxide (O2??). Using pharmacological inhibitors to lower O2?? production and breakdown, it was found that lowered O2?? levels resulted in a decrease in Pim-1 mediated Bad phosphorylation, a decrease in Bcl-xL expression and a NHE-2 dependent increase in Bax activation. Taken together, these results suggest a possible key role of O2?? in the PI3K-Akt independent survival pathway, providing LNCaP cells with an alternative means of evading apoptosis.
