EXPRESSION, FUNCTIONAL AND GENOMIC STUDY OF HEPARAN SULFATE 6-0-SULFOTRANSFERASE 3 IN BREAST CANCER. ScholarBank@NUS Repository. | Abstract: | Heparan sulfate 6-O-sulfation is biologically edited by 6-O-sulfotransferases (HS6STs) and Sulfates (SULFs) within heparan sulfate (HS). Three isoforms of HS6ST and two isoforms of SULF enzymes have been discovered in humans. These enzymes differentially expressed in a variety of human tissues and regulate the 6-O-sulfation within HS chain. Recently, several studies have proposed that dysregulation of 6-O-sulfation patterns in heparan sulfate chain may be involved in carcinogenesis. This study aimed to analyze the expression, functional and genomic role of HS6ST3 in breast cancer. HS6ST3 was silenced using siRNA and then functional studies were performed. Silencing HS6ST3 considerably diminished the tumor cell growth and progression. It was also modulated the cellular response to chemotherapeutic treatment. We introduced a novel pathway which critically regulates the tumoral cell behavior in breast cancer. In addition, HS6ST3 and SULF1 introduced as valuable prognostic biomarkers in breast cancer. | URI: | http://scholarbank.nus.edu.sg/handle/10635/33426 |
