Abstract: The allergen specific immunotherapy has been the treatment of choice against type I allergy. A new generation of oral vaccines is being developed using specific strains of lactobacilli. In our study, two Lactobacillus strains were genetically engineered to express a major house dust mite allergen by using an inducible pSIP expression system. The in vitro immunomodulatory effects of recombinant lactobacilli particularly on dendritic cells were characterized. Furthermore, their immunogenicity and prophylactic potential were evaluated in murine models. These findings on recombinant Lactobacillus strains will make valuable contributions in the development of safe and more efficacious Lactobacillus-based oral vaccine for the management and treatment of mite allergy in children.
