dc.description.abstract | Hydrogen sulphide (H2S) is endogenously produced in many mammalian systems. Recent studies suggest a possible role of H2S in the cardiovascular system and it was thus of interest to examine the cardiovascular effect(s) of H2S. H2S was a mild vasoconstrictor at low levels and a vasorelaxant at higher levels. Vasoconstriction possibly involved interaction with nitric oxide, whilst vasorelaxation was partly mediated by KB,ATP-channels in vitro but not in vivo. The (patho)physiological relevance of H2S was then investigated in haemorrhagic shock (HS). Hypotension, elevated plasma H2S levels and increased liver H2S biosynthesis was seen in HS and associated with higher levels of organ injury markers and pro-inflammatory cytokines. Inhibition of H2S biosynthesis with DL-propargylglycine (PAG) partially reversed HS-induced hypotension and was correlated with prolonged survival. PAG treatment before HS attenuated pro-inflammatory cytokine production and was protective in the liver and lung. PAG pre-treatment also modulated vasoconstrictor-induced changes in heart rate in HS. | |
