dc.description.abstract | Zebrafish mind bomb (mib) encodes a RING E3 ubiquitin ligase that promotes Delta ubiquitylation and endocytosis in Notch activation. A zebrafish mib homologue, mind bomb-like (mibL), was cloned. Mib and MibL can form homo- and hetero-oligomers and ubiquitylate themselves and each other. They function similarly in Notch signaling. However, MibL is different from Mib in the substrate specificity. I also revealed different severity among zebrafish mib alleles with mibta52b (M1013R) being the most severe, followed by mibm132 (C785stop) and mibtfi91 (Y60stop). Morpholino (MO) and mRNA injection data suggest that this order of severity is due to a dosage-dependent dominant-negative effect of Mibta52b and Mibm132 proteins on MibL. Furthermore, it was observed that Notch activity negatively regulates mib transcription in a Su(h)-dependent manner. | |
