Abstract: This thesis describes the roles of a germline organelle (nuage) and small RNAs in Drosophila melanogaster germline cells. First, I report that Maelstrom, a nuage component that localizes to both the nuage and the nucleus, has dual functions and can be regulated by its High Mobility Group box and phosphorylation. Maelstrom functions in the nucleus to repress the transcription of miRNA-7, which in turn represses differentiation factor Bag-of-marbles (Bam), thereby allowing germline stem cells to differentiate. Maelstrom also functions in the nuage to repress transposons via the piRNA pathway. Next, I show that another nuage component, Vasa, functions to promote mitotic chromosome condensation and segregation by facilitating robust chromosomal localization of condensin I Barren. Interestingly, the somatic paralog of Vasa, Belle, plays the same role in somatic cells, and it appears to be conserved in human cells. Therefore, RNAi components may play a conserved role in regulating condensin localization.
