Abstract: During the development of vertebrate hindbrain, the neural epithelium will be subdivided into a series of rhombomeres and a distinct population of boundary cells forms at the interface between each segment. Similar to the D/V boundary formation in the fly, Notch activation confers a boundary-type cell affinity and correlates with a delayed neurogenesis. Here, we provided further evidence that Notch activation was necessary for the boundary maintenance. Firstly, in mib mutant the cell morphology changes sequentially despite the correct initiation of hindbrain boundaries. Also, we noted that injecting su(h) morpholino into wild-type embryos led to a similar phenotype to that of mib and that different dosage resulted in different severity. Thus we propose the maintenance of hindbrain boundary is Notch-dependent. We further found that Notch1a and Notch5 are the receptors responsible for the boundary maintenance, since notch5 knock-down in notch1a mutants led to rhombomere boundary defects. Besides, we observed that glia formation and patterning were affected in mib or notch1a/notch5 knock-down mutants. These observations imply that in addition to the neurogenesis, Notch activation is required for gliogenesis in the hindbrain boundary region at later stages.
