ScholarBank@NUS Repository. | Abstract: | Mutations in the IRF6 (Interferon Regulatory Factor 6) gene caused Van der Woude syndrome (VWS) and popliteal pterygium syndrome (PPS), which are human autosomal dominant clefting disorders. However, the function of IRF6 remains unknown. In this project, molecular characterization and functional analysis of zebrafish irf6 was performed. The full length zebrafish irf6 cDNA, the genomic structure and the gene expression pattern were identified. The knockdown and R84C dominant-negative perturbation analyses were performed to observe the effect of reduction or loss of Irf6 function on organogenesis during zebrafish embryonic development. All loss of function treatments produced larvae with identical gross morphologies, including bloated abdomen, small eyes and head, markedly reduced liver, pancreas and intestine and the delay of the differentiation of epithelial cells.
