Abstract: Microdevices and miniaturized systems with biological and/or biochemical functionalities are regarded as one of the most important advancements for both research work and applications in biochemistry, biotechnology and medicine. However, in current microfabrication technology, there is no technique established for the integration of biomolecules with silicon/glass-based materials allowing further processing and fabrication of 3D microstructures. In addition, there is no satisfied biopatterning method which can fulfill the requirement of integration of biomolecules into the biopatterning process with desired patterning quality. This thesis work focuses on developing a technique for the biomolecule-integration in microfabrication as well as in biopatterning process. This was achieved by applying a protective gold film on biomolecules to protect them from degradation. The gold film protected biomolecules were successfully integrated into basic photolithography-based microfabrication processes and Focused Ion Beam based biopatterning process. In addition, the gold film protection was successfully applied in the dry storage of DNA on a compact disc at ambient temperature, allowing for the integrated storage of both digital information and biological materials. The developed technique may have applications in fabrication of various hybrids devices as well as in large scale biobanking systems.
