Abstract: While Vero E6 cells have been widely used in the study of the newly emerging severe acute respiratory syndrome, infection in human cell lines in vitro have not been established well. We have cloned and expressed the S glycoprotein of SARS-CoV in Vero E6 cells as well as a human cell line HLCZ. In this study, we analyzed the gene expression profile of cells that transiently expressed the Spike protein using microarray analysis. Our results showed that expression of the extracellular domain of the spike protein up-regulates the mRNA levels of 84 genes and the down regulation of 32 genes. The largest group of proteins, transcriptional factors and regulators were mostly up-regulated while genes encoding cell cytoskeleton proteins were generally down regulated. Our data demonstrates the changes in levels of expression of various genes, and identifies the intricate effects of a mock a??infectiona?? of a cell which provide new insights into the host pathogen interaction during an infection by SARS-CoV.
