Abstract: To assess the efficacy of cytokine gene therapy in bladder cancer, we transfected MB49 cells using DOTAP plus methyl-beta-cyclodextrin solubilized cholesterol as the delivery agent and TNF-alpha as the therapeutic gene. The results showed that MB49 cells produced 893.7A?24.0 pg/ml of TNF-alpha 2 days after transfection. Cell growth was inhibited and apoptosis was induced. MHC class I, B7.1 and Fas expression on the MB49 cells were increased. In vivo, an orthotopic murine bladder cancer model was used. TNF-alpha mRNA was increased after the first instillation and then returned to baseline 1 month after the sixth instillation. Three of twelve mice died from excessive tumor burdens in the control group while only 1/12 died in the TNF-alpha group. Histology revealed that 9/12 mice in the control group and 3/12 in the TNF-alpha group had bladder tumors. Our data indicates intravesical instillation of pBud-TNF-alpha produces a significant anti-tumor effect without severe toxicity.
