Abstract: Hypericin-PDT is a promising treatment modality for cancer. In this study, the effect of hypericin-PDT on NPC was investigated in vitro and in vivo. Well-differentiated HK1 NPC cell and poorly differentiated CNE-2 NPC cell were adopted in this study. Murine models were established by subcutaneous transplantation of NPC cells in Balb/c-nu/nu mice. It was found that Hypericin-PDT induced cell death in NPC cells was both cell type and dose dependent, in terms of apoptosis and necrosis. Hypericin phototoxicity was induced mainly through production of 1O2, though free radicals were also produced in this process. The generation of ROS resulted in lipid peroxidation and changed the activity of antioxidative enzyme, such as GST. Hypericin-PDT was also found to modulate the gene expression of GM-CSF, IL-6, MMP-1 and MMP-9. It is concluded that hypericin-PDT could effectively induce cell damage and tumor destruction, and decrease the metastatic potential of NPC.
