Abstract: Eubacterium coprostanoligenes has been found to be a cholesterol-reducing microorganism. The bacteria could have potential in cholesterol lowering conferred by its ability to convert cholesterol to coprostanol. The latter is poorly absorbed in human intestines and would be excreted. In our study, E. coprostanoligenes was cultured in Base Cholesterol Medium and its morphology was studied using confocal and transmission electron microscopy. Solid plate counting and gas chromatography were developed to monitor its growth and to investigate cholesterol reduction activity, respectively. It was found that growth and cholesterol reduction activity of the bacteria were affected by lecithin, CaCl2 and pH of culture medium. It was also found that growth of the bacteria was not affected by cholesterol substrate. In addition, anaerobic E. coprostanoligenes was observed to survive exposed to ambient air for at least 60 hours retaining its cholesterol-reducing ability. On the other hand, kinetics study of cholesterol reduction activity of the bacteria showed a Vmax of 14 I?M cholesterol reduced/hr and Km of 1 mM cholesterol. The secretion of the putative cholesterol-reducing enzyme(s) appeared to be constitutively and intracellularly. Lastly, an indirect pathway of cholesterol reduction was verified in E. coprostanoligenes
